Ya know, to be honest, I (Surreal) don't watch enough detective/investigation type shows to really have a wide range of choices. But when I read this question, someone did jump to my mind right away.
Megan Connor from The Sentinel, played by Anna Galvin. (Tin Man fans know her as Lavender Eyes or The Queen.)
She was awesome. An "exchange detective" from Australia in the final season of the series, she was brought in as a love interest for the boys. Problem is, by that point in the series The Boys were already firmly a couple. (Yeah, slasher here, but it's TRUE.) So instead, she became a unique and fun character of her own. At one point, she made a comment about how Jim and Blair, together, were the perfect man.
Plus, you can't go wrong with a woman who crawls into the duct system of the police station to WRESTLE AN ESCAPED CROCODILE. \o/
ETA: Anndie chiming in here. Again, I don't really watch that many, or at least don't pay attention to them. Will agree with Surr's choice of Megan. She rules. My other choice isn't an actual detective, but she works with them. I love Abbey from NCIS. She takes no crap from the boys, is a tried and true geek, plus is the go-to chick when they need something solved. Also, who can show up to work one day dressed as Marilyn Monroe and get away with it? She can.
